Next Generation Protein A Ligand

Co-developed with Navigo Proteins, NGL-Impact® A Ligand from Repligen was developed through a series of steps, including wild type Protein A shuffling, screening and multimerization.  The result is a market-leading ligand that responds to the need for capture resins with better binding capacity, caustic stability and elution while reducing costs and improving process economics.

Repligen and Navigo will co-develop a portfolio of next generation ligands, utilizing Navigo’s proprietary Precision Capturing® technology platform.

Commercialization

Repligen entered an agreement to commercialize NGL-Impact® A  Ligand using Purolite novel Praesto® agarose jetting base bead technology. 

NGL-Impact® A has outstanding performance attributes when immobilized on jetted agarose bead technology, with ultra-high binding capacities and excellent alkaline stability. These attributes are important in providing improved monoclonal antibody capture to handle increased upstream product titres, and in allowing higher concentrations of sodium hydroxide (NaOH) for sanitization of resins.

 

The following data show the performance of Praesto Jetted A50 resin from Purolite, which uses the NGL-Impact® A ligand, compared with standard industry capture resins.
