Solutions for Plasmid DNA (pDNA) Bioprocessing

Plasmid DNA (pDNA) is a critical raw material delivery template for the development and production of mRNA or viral vectors such as lentivirus and adeno-associated virus (AAV) vectors.

A large segment of viral vectors, critical in gene and cell therapy and other emerging modalities, are generated through transient expression routes based on plasmid DNA (pDNA) as a starting material. Ensuring adequate supplies of pDNA, made at increasingly larger manufacturing scales, is key to the development and delivery of these new therapeutic classes.

The fundamentals

What Is Plasmid DNA?

Plasmid DNA, or pDNA, is a small circular DNA molecule with an important job: carrying the genetic instructions that help enable many advanced therapeutic workflows. In biomanufacturing, pDNA often sits at the beginning of the process as a template, starting material, or critical input for the therapies that follow.

That early role makes pDNA especially important. It may be used as the DNA template for mRNA production, as an essential component in viral vector manufacturing, or as
the foundation for DNA vaccines, gene therapy, gene editing, and cell therapy development. In each case, the quality of the plasmid can influence how efficiently the next step performs.

For process development teams, pDNA is not just another intermediate. Yield, purity, plasmid form, impurity profile, and process consistency all matter because the plasmid carries forward into larger, more complex workflows. A strong pDNA process helps create a more reliable starting point for the therapies built from it
